The Doncaster Mile was first held in 1865 and Dundee won the inaugural race. Back then, the race was still held over eight furlongs or about 1600 metres. Also, the race back then is still a principal race. The race distance of the Doncaster Mile was adjusted to exactly 1600 metres in 1972 following the conversion of Australia to the metric system.

The race that year was won by Gunsynd. However, the race became a Group 1 race in 1980 with Iko taking home the title. In 1990 and 1991, Super Impose won the Doncaster Handicap and the Epsom Handicap for two consecutive years. It was Super Impose that made the Doncaster Handicap-Epsom Handicap double a viable bet for horse racing punters.

The Doncaster Handicap and the Epsom Handicap have the same race distance of 1600 metres and both are open handicap races. However, the Doncaster Handicap is the autumn equivalent of the Epsom Handicap and the Doncaster Mile is also part of the Sydney Autumn Racing Carnival together with the Golden Slipper Stakes, the Sydney Cup and the Rosehill Guineas.

The home turf of the Doncaster Mile is the Royal Randwick Racecourse which is the largest racetrack in New South Wales. Royal Randwick has a circumference of 2,227 metres and a home straight of 410 metres. Royal Randwick Racecourse features sweeping turns and an uphill slope on the home straight. Races at Royal Randwick are staged counter-clockwise.

Betting options for the Doncaster Mile can be categorized into standard bets and exotic bets. Classified under the standard bets are the win bet, the place bet and the each-way bet. Meanwhile, exotic bets include the trifecta bet, the quinella bet, the exacta bet and the first four bet. For the trifecta bet, you must pick the three race horses that will cross the Doncaster Mile finish line first in exact order.
